การเสริมฤทธิ์ต้านแบคทีเรียของ friedelin จากรักดำ (Diospyros curranii Merr.) ร่วมกับยาแอมพิซิลลินในการยับยั้งแบคทีเรียแกรมลบฉวยโอกาส

Abstract

            งานวิจัยนี้มีวัตถุประสงคเพื่อเปรียบเทียบประสิทธิภาพของสารบริสุทธิ์ friedelin จากสวนสกัดเฮกเซนของรากรักดํา ผสมกับยาปฏิชีวนะแอมพิซิลลิน ในการยับยั้งการเจริญของเชื้อแบคทีเรียแกรมลบฉวยโอกาส คือ Escherichia coli ATCC 25913, Acinetobacter baumannii และ Pseudomonas aeruginosa โดยหาคาดัชนีชี้วัดประสิทธิภาพรวม (FIC) ของสารบริสุทธิ์และยาปฏิชีวนะที่สามารถยับยั้งการเจริญของแบคทีเรียดวยวิธี Broth dilution susceptibility test ผลการทดลองแสดงใหเห็นวา คาความเขมขนที่ต่ำที่สุดที่สามารถยับยั้งการเจริญของเชื้อ 50% (minimal inhibitory concentration: MIC50) ของแอมพิซิลลินและ friedelin มีคาระหวาง 256.00-512.00 μM และ 512.00-1024.00 μM ตามลําดับ คาดัชนีชี้วัดประสิทธิภาพรวมของ friedelin รวมกับแอมพิซิลลิน ตอเชื้อทดสอบเทากับ 0.25-2.00 การวิเคราะหฤทธิ์รวมกันของสารทั้งสองชนิดเมื่อเทียบกับสารเพียงชนิดเดียว พบวา friedelin รวมกับแอมพิซิลลิน มีประสิทธิภาพการเสริมฤทธิ์ในการยับยั้ง E. coliATCC 25913 ไดดีกวาเชื้อทดสอบอื่น โดยมีคา FIC เทากับ 0.25 ผลการศึกษาครั้งนี้พิสูจนใหเห็นวา friedelin จากรากรักดํา สามารถเสริมฤทธิ์รวมกับแอมพิซิลลินในการยับยั้งเชื้อแกรมลบฉวยโอกาสไดอยางมีประสิทธิภาพ            The objective of this research was to compare synergistic effect of friedelin that isolated from hexane extract from the root of Diospyros curranii Merr., and ampicillin. The tests were conducted against opportunistic Gram-negative bacteria, Escherichia coli ATCC 25913, Acinetobacter baumannii and Pseudomonas aeruginosa. To determine value of fractional inhibitory concentration (FIC), the broth dilution susceptibility test was designed for detecting synergistic effect. The results were shown that MIC50 of ampicillin and friedelin against all bacterial solates were 256.00 to 512.00 μM and 512.00 to 1024.00 μM, respectively. The FICs of friedelin mixed with ampicillin were 0.25 to 2.00. Compared to individual drug, friedelin mixed with ampicillin was shown the best synergistic effect against E. coli ATCC 25913 with the FIC of 0.25. The results obtained here indicated that friedelin, isolated from the root of D. curranii, had an effective synergistic effect with ampicillin against opportunistic Gram-negative bacteria.
